The Ark of the Covenant is the centre point of Jewish worship and is the resting place of the Ten Commandments that Moses placed there. It is said to be a golden box. The Ark also contained a golden jar containing some of the manna from the Israelites trek in the wilderness , the rod of Aaron that budded. It is said that both of these items were removed some time before the construction of the first temple.
The story surrounding the Ark was that God promisesed to protect the Hebrews and in return they promised to obey his commands.

The temple was the crowning achievement of king solomons reign built in the capital city of Israel, Jerusalem. King David, his father, wanted to build the temple ealier as a perminant resting place for the Ark of the Covenant but David was forbidden by God to build a temple. "You will not build a house for My name," God said to David, "for you are a man of battles and have shed blood" (I Chronicles 28:3). Over 3,300 people were appointed to build the temple. The Ark of the Covenant was stored in a room known as the Holy of Holies. There was next to no furniture in the room and it was the most holy place in the temple, hence the name.
The Temple was used by the Israelites as their place of worship and attended communion there.
The ark was kept in the temple for many years until the temple was destroyed in 39BCE. In 463BCE the Babylonians began to give threats to Jerusalem and after taking over in 425BCE destroyed the temple. The babylonians were lead by nebuchadnezzer II, the king of Babylon at the time. After the destruction of the temple, the Ark was lost and has not been seen since.

There have been many myths that have surrounded the Ark of the Covenant, myths including, the God's spirit is present in the Ark and that who ever touches the Ark itself will immediately die as read in the bible verse (Num. 4:15): And when Aaron and his sons have finished covering the sanctuary and all the furnishings of the sanctuary, as the camp sets out, after that the sons of Kohath shall come to carry these, but they must not touch the holy things, lest they die. These are the things of the tent of meeting which the sons of Kohath are to carry.
As far as knowing what has become of the Ark, there are many legends and myths that the Ark has surfaced but all of the people claiming to have found the Ark are yet to give hard evidence that what they have found is the real Ark.
